 You can do that.  Settings - Screen - Screen Lock

 Just select the Use Custom Screenlock Command checkbox.  Oh, and type in the
command, of course.

The default Ctrl+Alt+l locks the screen, unless you want to change it.

Doesn't make sense to *unlock* screen with a command. To lock you have 
to enable the Dbus Extension module. Then you can use the command:

enlightenment_remote -desktop-lock

If you have the Dbus Extension module enabled and set DBUS_SESSION_BUS_ADDRESS
appropriattely in the console (ssh or something else) you can call
enlightenment_remote -desktop-lock
and it should work.

Settings-settings panel-screen-screen lock-advanced

At the bottom of the window you'll see 'use custom screenlock'

if you can get to the settings... you can just disable screenlock anwyay.. and
while you're at it you have access to everything.. rm -rf ~/ too...

I know you can configure E to use a different screen locking app, for 
example I use xscreensaver.  I don't know if you could put together a 
shell that includes your screen locking mechanism and your command to 
execute and put it there.  Likely, that only gets you half way though as 
the unlocking, you'd likely have to write your own wrapper for the 
locking mechanism.  Sorry, thinking out loud a bit. It's worth a try?
