[Histonet] p16 dilution

Hi Histonetters,
Does any of you use p16 Cintec from Roche/Ventana?  We wanted to dilute this 
antibody but wasn't sure how much to dilute it to.  Figured we could save a lot 
by doing so.  We have been staining our p16 with no current dilution.  Any 
suggestions will help.  If there are any other vendors that offer p16 other 
than Roche, that would be useful to know.

Hi, Laurie.  We use the Leica Bond and dispense it into an open vial for use on 
that instrument.  I suggest you contact the vendor for the IHC stainer you are 
using.  They should be able to tell you how to optimize the new Ventana Cintec 
P16 for use on your existing system.

[Histonet] P16 Antibody

We are looking to bring in P16 to do in house but are having a hard time 
getting the stain to be clean. We are having issues with positive staining on 
tissue that should be negative. We are working with a concentrated antibody 
that we have diluted out to 1:6000 (which is 6 times the amount suggested from 
vendors 1:200-1:1000). We might need to change clones or vendors and were 
wondering if you could tell us what kind of p16 (clone) you use for staining 
and possible what vendor you get it from? Any information or suggestions you 
have would greatly be appreciated. Thank you.

Good Morning Histonetters, I am wondering how all of you using p16 from Roche 
on a Leica staining system are going to adapt to the changes coming from Roche. 
 Are you going to pay the exorbitant price for the new kit and list the test as 
an LDT in your reports, or are you going to source one of the ASR antibodies 
out there and use that route?  I just want to be sure I can re work a research 
antibody and report it as an ASR test in our reports without CAP giving us a 
hard time about it.

We purchase the antibody from Ventana and run it on our Bond Max platforms.  
During re-optimization, we found we could dilute it 1:10 (15 minutes) following 
"H2" (high pH) retrieval and still get excellent results.

Ventana/Roche bought out MTM which had the patent, so they're the only vendor 
you can get it from.  Since you're using Bond you obviously don't want it in a 
dispenser, but you can buy their manual staining kit and just use the antibody 
from that.

Ventana/Roche bought out MTM which had the patent, so they're the only vendor 
you can get it from.  Since you're using Bond you obviously don't want it in a 
dispenser, but you can buy their manual staining kit and just use the antibody 
from that.

My company wants to do P16 but refuses to by any ventana products and I
have explained it is a waste of money to keep testing the antibodies from

Does anyone know of a comparable clone to the Ventana p16 clone E6H4?  I would 
prefer it to be undiluted (concentrate) so that we can titrate out the antibody 
ourselves.  We need it to have publications that would back this up and/or 
stained slide pictures I can show our pathologist.

Yes we monitor the temp and humidity. Retrieval is made fresh for every run.
We run the PT for the p16 in a water bath since we only run a few slides at
a time, but the water bath temp in also monitored. I should say that there
is some staining in these slides just not what the control usually looks
like. If we repeat the slides they do stain correctly. There is no
difference in the slide prep or the staining procedure. Any ideas.

I've been told that the HPV testing will be discontinued as of end of year. HPV 
expresses the P16 gene but so does other type of cancers associated with HPV.


It is possible to have a P16 pos and HPV negative test.
But is it not possible to have HPV pos and P16 neg.  if HPV is positive then 
the P16 is also positive.


So the P16 cannot be used to replace the HPV testing can it?

I was wondering if anyone would be willing to share a protocol for p16 
(Biogenix pre-dilute) antibody on the Benchmark XT.  I'm having a little

Now that MTM has made P16 very expensive and TriPath is have problems
producing ProExC, what's everybody using on ffpe tissue? We were told by
TriPath/BD that ProExC will probably not be available until late June.

Several of you have asked that I share our protocol for the P16 on the
Ventana XT.  I apologize for the delay, but here is what we have worked
up and it performs well in our lab:

CC1 Standard
Antibody incubation @ 37 degrees C. for 28 minutes
Amplification
Hematoxylin counterstain 4 minutes
Bluing Reagent 4 minutes

We place the commercial antibody from the CINtec kit in a prep kit and
use it as an RTU.

They hold the patent and no one else produces it.  We order the kit and use the 
antibody on the Ventana XT stainers, placing the antibody in a prep kit.  Works 
great.

Yes, it's true, only MTM labs sells it and they
Sell it as a kit and they also 
sell it along with a negative control reagent.
That's how I buy it.
They have a "license" or something and no else can sell it now.
Their cat# is 9518 and I think it's called the 
CINteck Histology V-kit,
(you should double check with them.)
They will make you "open up an account..."

If you want the IVD form of the antibody you have to go with MTM labs, they 
hold the patent.  We buy the kit and take out the antibody and use the 
detection for research.   It works very very well.

Hi Deanne,
My understanding is that you must get p16 from MTM Laboratories.  They
have some sort of monopoly right now. I still have some leftover from
Cell Marque and when I run low or the antibody is about to expire I must
also go to MTM.

I currently use the Cell Marque p16 Ready To Use by pretreating with
Dako's TRS for 40min in steamer, and detect with labelled polymer.
Dako's Envision + Mouse kit. It works nicely.
