[ccp4bb] A community support letter for the “UK XFEL” project

To whom it may concern,

To explore the UK and International community appetite for a next generation 
XFEL we are conducting this survey to gather information and sample support for 
the UK XFEL project (which seeks to deliver such next generation capability).

X-ray free electron lasers (XFELS) have emerged as important and wide-ranging 
tools for advanced science over the last decade. Their impact has been 
extensive (from catalysis and energy research to structural biology and 
laboratory astrophysics) and has opened-up a new era in research in structural 
dynamics. In the UK and around the world significant numbers of scientists have 
begun to engage with these opportunities, and many more are hoping to do so as 
the capabilities are developed, and as capacity grows so that it becomes easier 
to secure access.
The UK XFEL Science Case

examines the transformative scientific opportunities that will be created with 
a Next Generation XFEL i.e. one that can deliver near transform limited pulses, 
high reproducibility, and synchronised to a wide range of external beams (e.g. 
lasers, THz and electrons) all delivered at a high repetition rate with a pulse 
structure matching state-of-the-art sample delivery and detector technologies. 
We strongly endorse this scientific vision, and fully support the next stage in 
the efforts to ensure that there is an overall capacity increase and that these 
new capabilities can be made available over the next decade for advancing 
science and technology.

[ccp4bb] Post-doctoral position available

The Page Laboratory at the University of Connecticut Health Center has an 
immediate opening for up to two post-doctoral candidates to study pro- and 
eukaryotic signaling enzymes, with a focus on the function of phosphatases that 
form large signaling complexes, and also drug/inhibitor development. All 
studies use X-ray crystallography and/or cryo-EM/NMR. Applicants with a strong 
background in biochemical techniques, and especially eukaryotic protein 
expression and protein crystallization, are encouraged to apply.


The Page laboratory is housed in a newly renovated ~250,000 sq. ft. research 
laboratory in Farmington, CT. The laboratories are open-lab-space facilities 
and allow close interaction with the adjunct structural biology and 
molecular/cell biology groups at UConn Health. The Page laboratory is equipped 
with all necessary infrastructure to produce (E. coli, Sf9, mammalian), purify 
and analyze protein and protein complexes (ITC, SPR etc.) and crystallization 
robotics for crystal production.


Farmington, CT, is located on the New England between Boston (90 min.) and New 
York City (90 min.). Starting date, salary and project details are negotiable.

crystal.

I have also seen a talk where someone successfully dissolved their 
water-insoluble ligand in Octan-1-ol, and layered that solution over the drop 
after crystals had grown. They incubated for X period of time (I don't recall) 
froze the crystals and found the ligand had diffused into the crystals and was 
in the active site.

Maybe someone else knows of the/a paper describing this?

Hello everyone,

I am finding it difficult for getting a ligand density inside the protein as 
the ligand is very much insoluble. It's only soluble in 100% DMSO. I tried for 
co crystalization. Kd value of the ligand is near 40um. Any suggestion what to 
do?

Dear Gourab,
You might just want to add the ligand in pure, solid form to the edge of the 
crystallization drop. This worked in our case beautifully (see Angew. Chem. 
Int. Ed. 2012, 51, 3354). We had to soak it for 4 weeks.
