[ccp4bb] X-ray system available

2018-08-10 Thread McEwan, Paul
Dear CCP4bb readers,
We are in the process of decommissioning our in-house X-ray source and would
like to offer it to the community. The device is showing its age based on the
technology, but hopefully somebody could make use of it. In brief, the
equipment is a Rigaku 007-HF generator with two Raxis4++ image plate detectors
and computers for each of the ports. It is also equipped with a water to air
chiller system to cool the anode.

If you are interested please drop myself and John Barker an email:
paul.mce...@evotec.com
john.bar...@evotec.com

I would note that Evotec offers this equipment on an as is basis and interested
parties would be responsible for transportation and associated costs.

Best wishes,

Hi Francesca,
You could try using reductive methylation, this has always lead to a loss of
solubility (and protein!) in my experience. Jena bioscience and Hampton

Hi Francesca,

Try some zinc (1mM + ) in your protein buffer? Zinc tends to make a lot of

Hi all,

I am struggling with a protein complex that is too soluble. I have reached

and going to higher concentration is not easily achievable.
I have tried different buffer conditions (salt concentration and pH) and I am
testing temperatures. I am at a loss with what to try next.
Do you think PTMs (phosphorylation, acetylation) might be causing this?
Any input on how to decrease solubility?

Thank you very much in advance,

Rather than looking at anti-DTT it is more important to set up an
appropriate redox system. This can be a combination of reduced and
oxidised glutathione or cysteine. If you check some of the commercial
protein refolding screens this should give you an idea about relative
concentrations.

Use the 15 minute protocol from the Hampton Research catalogue/website. They're normally quite small and easy to cryoprtect with a little glycerol. Size can also be varied by playing with the salt concentration (slightly higher [NaCl]giving more but smaller xtals)
