Aloha Histonetters--I've missed you!

  1.  It's been a very long time since I last posted here so please forgive my 
heap of ignorance: I haven't done this in a hot minute.
  2.  I think I'm in the market for a cryostat! But it's been 20+ years since 
the last one I bought, and I have no clue what features I'll need now. Do I buy 
a reliable 90s analog model? Do I need digital control? No idea! I'd love to 
talk to sales reps to get the pitch...but because I'm most likely to buy a used 
model, I feel like I'd be wasting their time. So I thought I'd ask you fine 
people.

TL;DR: What features would you recommend are very useful for a cryostat for a 
low-volume research lab? And is it a bad idea to expect to do H and other 
stains on cryosectioned tissue?

To get the best advice, I'll try to anticipate some of the more likely 
questions and answer:
 "What do you want to do?"
Ideally, we'd like to do most of the same staining that one would do on FFPE 
tissue (H and some other stains) on fixed or unfixed frozen tissue. However 
more importantly I anticipate a lot of IHC/IF, and probably most important to 
us is preserving lipids in sectioned tissue (hence avoiding the clearing agents 
used with paraffin).
"What kind of tissue?" We're a tissue engineering company working extensively 
with mammary cells, so the workload will likely be fatty tissue, and some 
composite samples that are tissue and polymer scaffolds (nothing a good blade 
can't easily cut--I think).
"What volume?" Pretty low. I may well be underestimating the workload, but most 
likely a dozen blocks every few weeks.
"What budget?" That what I need help with. I can probably go up to $20k, and 
mgmt is OK with a new model if it's necessary, but frankly I wasn't fantastic 
on a cryostat the last time I bought one so I'm not confident I'm qualified to 
say what exactly we need.
"What is your skill level?" Noob. I've used a cryostat before, but my 
technique...definitely needs improvement. On the other hand, I'm the only one 
in my company that has any experience with histology at all, so I anticipate 
training a new generation of research histologists. So, I probably want a 
cryostat that may not do everything for me, but also won't require a lot of 
shoehorning.

I'll also leave a more open question down here: What is a paraffin microtome 
better suited to than a cryostat, and vice-versa?

We have 3 new leicas and we constantly have to adjust them. They creep forward 
so much that I just automatically know to roll it slightly back after every 4 
specimens or so. The new leica designs are just bad and customer service is 
even worse. We have to use the plate as we do Mohs specimens and the sections 
are just better with the glass plate. I want to look for a different machine 
next time. We did test the Avantik and it cut nice but the glass plate was on 
the front and got in the way. I wish they would talk to Mohs techs before they 
design these. Oh well. No tricks just constant adjusting. Thanks Anne

the roll glass helps, but it is still seems a lot easier for us to use a brush 
or a teasing needle.

I use roll plate. 

I don't like the roll plate on the Leica. The Leica design creeps... a lot. 

Also, with the leica, the contact point with the blade is a soft aluminum arm. 
If you make adjustments with the roll plate holder contacting the blade, it 
will damage the soft aluminum holder. 

I like the thermo design much better. It creeps a lot less. 

Here's a procedure for roll plate adjustment. 

1.  Mount block on specimen disc. 
2.  Place Specimen Disc in chuck, position Blade carrier, and face off.
3.  Turn knurling screw clockwise until sections do not pass under the 
Anti-roll plate.
4.  Turn knurling screw counter-clockwise by 20 degree increments until 
sections effortlessly pass under the Anti-Roll Plate WITHOUT hitting the block. 

*   If the block is hitting the anti-roll plate, it may cause damage to 
glass. 
*   Hard contact is visible and can be heard. 
*   Soft contact, which can also damage the Anti-roll plate, cannot be seen 
or heard. But, can be felt by gently placing finger on the anti-roll plate 
turning lever while taking a section.

[Histonet] Cryostat cutting head temperatures

I was asking about the cutting head temperature because the Leica CM3050 
literature says the cutting head can be adjusted down to -50c separately from 
the chamber temperature. Does anyone out there use this unit? It seems to be 
very popular with the research community.  Do you ever adjust the temperature 
of the cutting head down that low, or do you just adjust the chamber 
temperature? Why would you ever use this separate feature?  Why would you chose 
this unit over the 1850?

froze it, cut and stained duplicate blocks and had the pathologist sign off on 
the process.  We were also able to get some tonsils delivered fresh and did

longer available which could be,. like she said, years. We also have a
local company do the PM and repairs and will use it until
it absolutely cannot be repaired.

I'm not sure I understand why you would want to replace your 1850s just because 
Leica is closing the support.  I have 2 and they are real workhorses.  There 
are 2 independent companies in the area that can repair and perform PMs.  One 
is Bel-Air and the other is Advantik.  I'm sure there are others, but I've used 
both companies and can highly recommend them.  If you like your Leica, keep it 
until parts are no longer available, which could be years.

Hi Brittany,
Personally I do not like the HM525NX.  It is awkward and difficult to control 
with no manual advance except the control panel.  When advancing /trimming you 
are never certain how far forward the block will travel.  The UV light fitting 
in the lid always seems to get in the way of my field of view.  Ours doesn't 
have temperature control of the block holder which can make sectioning more 
difficult.

Looking at the specs of the cryostar I would actually opt for the NX70.

will probably be available in the states.

purchase (e.g. height adjustment, vacutome, etc.)?

Guess I missed your other questions. 

I think the fog mist (Cold D) may be better - it reaches all area of the 
chamber. UV light is shaded from some areas, though probably hits most areas 
you might be touching. 

The misting requires a specific solution, not just anything. Hydrogen peroxide 
is the disinfecting agent. 

Other features:
We have the vacutome on the original units but we never use it. We do all 
kidney and muscle sectioning and these small samples do not produce much 
debris. On our new replacement unit (to replace a 7-year old NX70) we opted out 
of the vacuum.  If using in a gross room/Or setting the vacuum may be very 
useful. It only adds a hose and suction manifold in the unit and does not get 
in the way of anything.

[Histonet] Cryostat question

Does anyone out there use the Avantik QS12 cryostats? We have a machine that is 
only a year and a half old and have had to get the Peltier system (the cryobar) 
replaced 4 times. Is anyone else having this problem consistently?

[Histonet] Cryostat and Parts

We are in the process of moving and we have some equipment and supplies
that we are not taking with us.We have 2 Microm HM 505E cryostats and
all of the internal components.One of the cryostats doesn't cool down
and the other one doesn't advance properly but is otherwise in working
order.If anyone is interested in the cryostats or the any of the parts
please let me know.We also have 12 bottles of Polarstat Plus cryostat
mounting media (exp 6/18).If anyone is interested please let me know.

Our last discussion with TJC to clarify their definition of "regular interval" 
was to follow the manufacturers guidelines.  I recommend determining what 
manufacturer suggests with the use of the UV decon and write that in your 
policy that addresses this standard.  If you are following their guidelines, 
you should be in compliance.

[Histonet] Cryostat sectioning of frozen leaves

I am trying to cut frozen leaf sections at 6 and 8 microns.  All I am getting 
is bunched up leaf.  What is the trick/  I am cutting at -20 should be cutting 
at a lower temp and cutting thicker?

[Histonet] Cryostat decontamination ANP23410

Hi Everyone,
Just a quick question. If you defrost your cryostat to room temperature and 
wipe it out with 70% alcohol is that considered decontamination? Or does it 
have to be fumigated with formaldehyde, or have a UV function on board the 
machine?

I think that might have to do with the fan motor inside the unit, it might need 
to be replaced.   We had ours replaced a few years ago.  Do you service/PM the

Our Mohs Lab is CAP accredited. Every seven  days of use, cryostats are shut 
down. Once machine has been cleaned and dried we use Cavi-wipes(super 
Sani-Cloth from PDI Inc.)  We then document disinfection date on QC Log.   We 
have three cryostats, we rotate a cryostat a week for disinfecting purposes. We 
added this to our Equipment: Cryostats/Microtomes policy.

[Histonet] Cryostat sectioning of FF lymph node

Good morning!  My pathologists would like us to cut formalin fixed (not yet 
processed) tonsil tissue on a cryostat for DIF staining.  Has anyone done this? 
 I did a quick search that seemed to indicate that it was possible but that the 
architecture of the cells would be altered because of ice crystals and that it 
would be difficult to get the sections to stick.  If you have any advice I 
would greatly appreciate it!

[Histonet] Cryostat purchasing

Hi Everyone,

Can anyone recommend a good Cryostat to buy for use with Human Tissues.

I'd like to look for a reasonably priced model.
Also, I'd consider paying extra for a model that is designed for human tissues 
so it has disinfection ability or is easier to disinfect than older models.
I'd also consider going with a refurbished model if the price was right.

I am used to cutting on CM3000 and CM3050s which are quite old.

[Histonet] Cryostat troublshooting. CM3050

H everyone,

We have been using this old CM3050 cryostat for our Frozen OCT blocks.

I am cutting at 5 uM.

My issue is that when I use the hand rotator to cut my sections it doesn't seem 
to advance with each rotation.  It can take as many as 4 rotations to go from 
one section to another.

What would cause this, and is there an easy solution to this problem?

I can use the button to automatically move the chick holder all the forward and 
back from blinking to blinking, so I am wondering if ice crystals or debris can 
be ruled out as  the issue.

Any advice welcome.

Hello Patrick,

We have this same cryostat except it's the CM 3050S.  Ours also has
this issue usually after it's been running for many months with a
defrost.  The typical cause of your problem is the cylinder on which
the block holder is attached has ice crystals or is frozen.  I feel
the cause of this is because the glass was left open too long while
cutting.  This allowed moister from the room to condense on the
cylinder and subsequently causes a stop go motion.

 Since I am in charge of all histology equipment maintenance, my
solution is to turn off the cryostat, take the microtome out of the
cavity and let it dry overnight.  Then in the morning, I physically
dry the microtome.  Using cryostat oil, oil all the moving parts
including the cylinder, then wipe off excess.  Before putting the
microtome back into the cavity, the cavity itself must also be
thoroughly dried.  The final cool down process usually takes about 1-2
hours.

Are you required to revalidate all of your IHC antibodies once you switch to a 
xylene sub?

We had utilized a xylene free process in our Peloris, which utilized alcohol 
but we ran into issue with FISH for Her2 and some of our other predictive 
markers and abandoned the alcohol.

Instead of xylene you can use isopropyl (also known as 2-propanol and propyl 
alcohol) and after sectioning you can use dishwasher soap before going directly 
to the staining procedure. After staining you need nothing but an oven to dry 
out the stained sections and go diractly to coverslipping.

Hello all;

Is anyone aware of a cryostat design in which the microtome cabinet
would sit at bench level?  In other words, it would function like any
regular bench microtome but be housed in a cold cabinet like a cryostat?
This would enable the operator to work in a desk position instead of
uncomfortably being hunched over a cryostat cabinet.

Dear Kim,

Yes the Starlet cryostat made by Bright Instrument Company, it is in a small 
bench level cabinet as there is no other way of achieving good results on the 
refrigeration. Apart from freezing microtones which were highly inefficient on 
retrieving good quality thin sections.

[Histonet] Cryostat Disinfection

In the CAP checklist it states that The cryostat must be4 defrosted and 
decontaminated by wiping all exposed surfaces with a tuberculocidal 
disinfectant.  The cryostat should be at room temperature during 
decontamination unless otherwise specified by the manufacturer.   The newest 
cryostat by Thermofisher uses cold disinfection with a chemical called Sanosil. 
  Am I right thinking that on that cryostat we do not have to disinfect at room 
temperature?

Tanya and Madeline,

Jan Minshew of Leica sent a nicely detailed response to Decontaminating a 
cryostat in 
January of 2010. She apparently wrote it to put in a Leica How To manual, 
although I did
not notice it in this form.  Everything she states is well documented, although 
I also 
incorporate a known virucide and tuberculocidal product (following it's 
directions, naturally).

If you have a UV cryostat, use Leica's Cryofect product or follow Tim Morken's 
previous
(2000 histosearch)formaldehyde method, they should work for killing most bugs 
too(with the 
exception of eradicating some prions like CJD/BSE).


If I am misunderstanding what you mean by Decon I apologize, as there is no 
reason why
you cannot clean up tissue debris in a cold cryostat with 100% alcohol.

[Histonet] cryostat

Does anyone have any current information on the cryostats with UV for 
decontamination?  Specifically, is it accepted as a means of decontamination by 
CAP or JS or any other regulatory agencies?  Basically, is it worth the extra 
dollars to go that route?

The only method acceptable to CAP is bringing the cryostat to room temp and 
disinfecting weekly UV light will not be an acceptable alternative by CAP.
We clean out all tissue debris, wipe down with abs alcohol, bring cryostat to 
room temp, use Cavicide to disinfect, wipe down again with abs alcohol. We use 
70 % alcohol for daily cleaning.

There are many tuberculocidal cleaners. Just search for it. If you are in a 
hospital the central supply will most likely have some in stock.  
We use PD-128 by Spartan Chemical

effective against HIV).

Recommends formalin fuming for 24-48 hours at room temperature. Followed by 
ammonia for one hour (presumably as a neutralizer).

Recommends defrosting , washing with 70% alcohol, complete cleaning. (no 
references given).

We wipe out the excess tisue( if there is any),  wipe down the chamber with 
abs. Alcohol then spray the chamber with CaviCide, let it sit for @ 3-5 
minutes and then wipe the chamber out again.

[Histonet] Cryostat recommendations

Happy Friday everyone!
I'm looking for some feedback on cryostat preferences. We are replacing one of 
ours now and will be evaluating a few before making a final decision. The 2 
I've looked into so far are the Leica 1860 and the HM525 by Thermo Scientific. 
I'm interested in the UV feature that many now offer too and want to know what 
your experiences are. Does it require extra maintenance? Is it costly to 
repair/replace?  Do either of these models (or previous versions) have any 
known problems? They both look very nice on the outside, but are they made of 
durable materials internally? Thanks in advance for your responses, and have a

[Histonet] cryostat decontamination

Has anyone come up with a documented and referenced procedure for
decontamination of a cryostat using 37% Formaldehyde?  

 

I pulled up old notes in the Histonet archives and saw that Tim Morken
was working on this but that was a number of years ago.

I would not vacuum the trimmings out of cryostat, if it were infected It would 
make a good job of contaminating a laboratory etc.

There are vacumes specifically made for cryostats that are hepa filtered, etc.  
We have one of those.  The company is called MARMED.  We use a shop vac for the 
paraffin microtomes, that works nicely.

Hi Irena!

If you go to IMEB Inc.'s website, and type vacuum into the search, it will 
take you to what we use.  They sell a vacuum cleaner and accessories.  We 
currently use the filtered hoses with an old canister vacuum that's been around 
longer than I have.  When the filter gets full, we discard it in the biohazard 
trash and replace it with a new one.

[Histonet] CRYOSTAT PROBLEM

Hi all,
We have a old cryostat (Microm h505 model). It was making some sudden 
vibrating noises for a little while and then smooths it self. There  are no 
temperature alterations every thing was fine but suddenly it stopped working, 
no display more looks like dead :( 
I tried calling the service company which serviced before but I guess the 
company is no more in service (as it was a very old machine). I am very new to 
this maintenance issues , I would be glad if any one of you can help me in 
suggesting a company or person whom I can talk about the issue or some one who 
can come and look at the  instrument and suggest the best advice . I assume 
probably some electric motor is dead inside. 
Any advice will be really helpful.

[Histonet] Cryostat cutting?

Hello,

We need to cut clean and consistent 50 micron sections of OCT embedded
tissue on our leica cryostat (CM-1850). Does anyone on the list have
any tips or tricks for doing this when compared to cutting the usual
10 micron sections that we usually deal with in our lab.

Mikron Instruments services most of our instruments, including Micron
brand.  I don't have any Leica equipment now, but they have serviced my
Leica microtome in the past.  They are very good!  The service manager's

The compressor fan was loose, so when the Tech One guy fixed it, instead of
making a horrible noise when it turns off, it just rattles.  Oh well, time
for a new motor.
Which leads me to another question, is there any reason I should call Lecia
to replace this motor? The guy from Tech One Biomedical seemed to know his
stuff, but my boss seems to think we should call the people who made the
cryostat.  Personally, I think that would be spending a lot more money for
the same thing, but maybe I'm missing something.  Any suggestions?
By the way, the cryostat is from 2008 and isn't under warranty.

Our cryostat (CM3050S) is making a terrible noise when the fan stops
running, yet I don't want to call Leica (the manufacturer) because they
charge more money than anyone could ever afford.  How would I go about
finding someone who could repair a cryostat in Pittsburgh? I don't think
it's the refrigeration that's broken, it sounds like something mechanical.
Any suggestions?
Has anyone ever tried to open the back of one to see if it's fixable? I'm no
mechanic, but I think I can spot a worn belt or loose screw.  I'm just not
sure I want to open it up myself.

You can try Ray Brodersen at Brodersen Instrument Co, they are in Mars
and do most of our repairs. Ray does an excellent job at a good price.
It sounds like the bearings in your fan or maybe the compressor.

Dear Histonetters,

Our 11 year old cryostat is beginning to experience some unusual problems. Our 
Medical Maintenance says that they won't put a lot of money into fixing it 
because of the age of the unit. Therefore, I am asking for opinions, pros, cons 
of different brands of cryostats. I am interested in a cryostat that has the UV 
light or other source of easy decontamination. We don't do a lot of frozen 
sections but enough for us to have to have a good cryostat that is easy to

  We have been happy with the Leica cryostats over the years. We have an 1800 
that is about 15 years old, an 1850 that is about 5 years old with the UV light 
for decontamination.

Medical Maintenance says that they won't put a lot of money into fixing it 
because of the age of the unit. Therefore, I am asking for opinions, pros, cons 
of different brands of cryostats. I am interested in a cryostat that has the UV 
light or other source of easy decontamination. We don't do a lot of frozen 
sections but enough for us to have to have a good cryostat that is easy to 
operate and clean.  Any comments or suggestions would be greatly appreciated.

That depends on what staining you need to use.
If an IHC, just fix the sections in acetone, air dry and proceed without HIER
If HE fix quickly with formalin 10%, wash → hematoxylin for 30 secs without 
differentiation → eosin 15 sec→ dehydrate and mount.
Other HC procedures all will require fixation and using the specific protocol.
Always remember that it is a frozen section and therefore more delicate and 
less adhered than a paraffin section.

We have a similar frozen section protocol but we fix the slide in 95% alcohol 
for ~1minute and then stain with Gill's Hematoxylin for 1 minute, rinse, blue 
and then alcohol, foloowed by Eosin for ~ 15secs.- dehydrate and mount.

We have this model as well and like it.  We got the vacuum attachment.  It's not
perfect, but does facilitate cleaning up OCT and tissue scraps.

many used for Mohs, as holding a low temperature is not a problem. I can email

cryostat?  Can the specimen head be manipulated to acccomodate a badly embedded 
block? Does the machine stay cold enough to perform Mohs? Any advice would be

It may help to let your block equilibrate for about 20 minutes in the
cryostat.  I've found blocks do not section well if you start sectioning
right away, especially snap frozen ones.  Also, do you embed in OCT/sucrose?
A 1:1 OCT: 30% sucrose solution is much softer than just OCT (or whatever
embedding medium you are using).
What do you mean by the tissue is condensed? It's folding up? I've found
sectioning fresh unfixed tissue on a cryostat is impossible (we tried chick
embryo trunks, which may be harder to section fresh than other tissue)--it
doesn't section but get smushed to a pulp.  You need to use a sledge
microtome, unfortunately.
What thickness are you sectioning at? I've found sectioning above sixty
microns doesn't work as well on a cryostat, but again this is on fixed chick
embryo tissue.
If your cryostat is old, I would suggest you look for a new anti-roll plate
made of glass.  They work so much better.  As long as your cryostat isn't
older than about ten years, you should be able to find one that fits your
knife holder very easily.
That said, I've used a Rei-something cryostat that was 15 years old and it
sucked.  The anti-roll plate holder was too loose to do anything useful and
the adjustment of the blade holder was impossible.  Can you use a newer
cryostat in your department from another lab? Or just go back to sledge
microtome sectioning--if you know how to do it, it's the same principle
anyway.

Our Hospital has several Frozen Section Rooms, each of which have 1 to 2 
cryostats.
The majority of our cryostats are the newer models, with the built in 
Ultraviolet Radiation Disinfection cycle (runs every afternoon.)  However, we 
still have a couple of the older models that do not have an automatic 
disinfection cycle.What are other institutions currently doing to disinfect 
their cryostats.

Hy Karyrn.

As my lab carry frozen section daily, the decontamination/fumigation of the
cryostat on a daily basis is not practical as we do not have a back up
cryostat. Also after every frozen section, we make sure that the Cryostat is
get disinfected of any fresh tissue trimming with 70 % alcohol.
Every friday night the cryostat get set-up for fumigation and get
decontaminated weekly using Formaldehyde vapour/ then we neutralise them
with ammonia, according to manufacturer recommendation (Thermo Scientific),
though the fumigation will not activate until Sunday night so Week-end
On-Call staff can carry urgent Frozen sections if needed.  Each
decontamination/fumigation cycle is then Log, for CPA purposes.

Hello,
Our Hospital has several Frozen Section Rooms, each of which have 1 to 2
cryostats.
The majority of our cryostats are the newer models, with the built in
Ultraviolet Radiation Disinfection cycle (runs every afternoon.)  However,
we still have a couple of the older models that do not have an automatic
disinfection cycle.What are other institutions currently doing to
disinfect their cryostats.

[Histonet] Cryostat vs decal sections of bone

Hi all,

last year sometime I was asked to budget for some equipment for our unit.
Not expecting to get anything i aimed high, and requested a cryostat and
cryoJane system.
Lo and behold, the planetary influences were just right, and my request was
approved. Now, I've got cold feet (pardon the pun) and I'm wondering if
there *are*  any distinct advantages of cryosections of bone over
demineralised wax embedded samples in regards to immuno and in-situ ?.
