uploading or is there some software you use to do it all at once?
To do it best you should resize your image to the dimensions you want
and then after that, sharpen to taste for that size. Then save it

As was stated by someone else, for the web, you don't really need to
worry about DPI, just the total number of pixels. I'm not sure what
software you are using to do you sizing/sharpening - but I just set
the final pixel size, and then sharpen. If you save that as a tiff,
you don't need to worry
