If they're both up-to-date it should also just work, but it may not be  
obvious which one it will use (that's an implementation detail that I  
even should look up). However, in any case, synctex is much better in  
many ways, you simply should not use pdfsync anymore.

I don't really think it's the case sensitivity of the file system per  
se, it also depends on the (tex) program, I think. At least, for me  
PDFSync does not work with your file even though I have a case  
insensitive file system.

Indeed, Skim just does nothing. For this case I really think an alert  
is annoying, and even a beep doesn't work well. One problem is that  
the sync works delayed (because the initial parsing is relatively  
slow). And a beep or alert that shows after some delay is just  
confusing, because it has no clear relation to the event.
