Nicholas,

It may depend upon what version you are using. I recently upgraded my
RedHat 7 system to 7.2 and the resulting Wine was broken. Used rpm to
remove then reinstall. I'm not sure what the old version of Wine was,
but it used wine.conf. The new one is Wine 20010822 - it requires a
configuration file in ~/.wine/conf instead of the old /etc/wine/wine.conf.

Also check the documentation: on my system
/usr/share/doc/wine-20010822/. There are some helpful sample files
and also good information on dealing with vfat partitions so that you
don't need to run as root.

Cheers,
Alan
