I suspect the official Lion USB boot disk will be slightly different to the 3.4GB installesd.dmg file. You can't upgrade a Mac from 10.5 to 10.7 using the Apple Store downloaded version, even if you boot from an external drive. You have to install 10.6 first.
The official USB disk will probably be a full retail copy for those who haven't yet upgraded to SL.
