Mimedefang uses sa-mimedefang.cf rather than local.cf.
That's not what's biting him based on reading his message.
I think the original poster is attempting to change subject-line tags by modifying his local.cf. That will not work in MimeDefang, as you need to edit your MimeDefang configs to do this.
Mimedefang uses SA, and SA called by MimeDefang will use local.cf if sa-mimedefang.cf isn't present.

But based on the post, that's the prefs file, not the site rules.. seems quite strange to use local.cf as a prefs file, as SA will parse /etc/mail/spamassassin/*.cf as the site rules anyway. To double-specify the file is IMO silly and a waste of CPU time, but I may be getting the wrong impression.
HOWEVER, Mimedefang generates it's own message markups, so any subject-line changes, body reports, etc that are generated by SA will not be used. This is true no matter what SA configfile you use. You can change the subject tag in sa-mimedefang.cf until you're blue in the face and it won't change anything about the email.
