Short answer: yes you can. 

Long answer: yes you can - you can even use compression. There are two options: 

1) Using ZFS as file server (NAS)
2) Using ZFS as backend iSCSI storage backend for your Windows fileserver (SAN) 

Both is possible, while I would prefer 2) if you use Windows AD. 

In both cases make sure you have enouth RAM or L2ARC SSD in your box. 

Estimate: for 5 TB you would need between 10 GB (128kb recordsize) and 125 GB 
(8k recordsize) (depending on blocksize). 

Suggestion: 12 GB of Main Memory + 2 x 80 GB SSD for L2ARC (this way you can 
even go with 8 gb records size).
