This isn't happening when you are booted off a CD or other read-only medium,
is it?  That would keep Stickies from being able to save it's notes, since
it saves them in the active system folder.  When you're booted off a CD, the
active system folder is obviously read-only.

Another thing I'd try is moving the Stickies preferences file out of the
preferences folder and let Stickies make a new one, see if that fixes the
problem.

You could copy the text of your sticky notes into SimpleText or something
else so you don't loose them when you delete the (possibly) corrupted
preferences file.
