It's a button not a check box. It says "Ask where to save each file before downloading" followed by only button if it's not pressed and button pressed if it is. I just opened Chrome and went to it. It said pressed. I pressed the space bar and it said nothing. But I tabbed off and back onto it and it just says the name and button. I closed the window with Control+W and then closed Chrome with the same. I opened it again and it was still not pressed. I then did the same in reverse to turn it back on and now it says button pressed when I tab onto it.

On a link you can open the context menu and press K for Save link as.
Otherwise, here's how to set the global download options.
1. Press Alt to focus the Chrome button and the space bar to open the
context menu.
2. Press S for settings.
3. Tab down to the Advanced button and press the space bar.
Turn on browse mode and press H to move through the settings by heading.
Under the downloads heading you should see the default folder followed by an
unlabelled button. You can press that button to choose a folder.
You can also skip that and go to the next button which is to tell Chrome to
ask you where to download every time you download something.
