One possibility:

At some point in the past, you probably got a warning message that
read:

A script from "file://" is requesting enhanced abilities that are
UNSAFE and could be used to compromise your machine or data...

If so, and you responded to this message with "deny" and also selected
the [x]remember this decision option, then you've basically told
FireFox to always block TiddlyWiki's attempts to save to your local
filesystem which would, of course, trigger the display of the "Failed
to save..." message that you are reporting.

has some very good information and an excellent plugin that you can
use to manage your Firefox privileges on a per-file basis, which will
allow you to enable file saving for each specific TiddlyWiki document
while still blocking general file I/O permissions for other locally-
loaded HTML files.
