For the horizontal menue I'd go with NestedSliderPlugin [1] because it
allready does most of it. Some additional CSS is needed for the
<<list>> macro inside the second menue eg: .floatingPanel ul {}

For the layout you could go with emastic framework [2], deviding it
into rows, columns and boxes.
rows are containing columns
columns define the width and are containing boxes
boxes hold the content(tiddlers) and defines margin and or padding

1) row .. header dp100 full size empty. Only Backstage button could be
there
2) row .. container either fixed px size or eg: 85% centered contains
the rest of the page
inside 2)
3) column leftmenue dp25 which means 25% of container width. Contains
TW MainMenu tiddler
4) column banner + TopMenu dp75; 75% of container width
inside 4
4.1 box BannerTiddler contains the big banner
4.2 box TopMenue tiddler contains nestedSlider see above
5) column TW ViewTemplate dp50 50% of container
inside 5) if you need the 3 pics below the content
3 boxes each 33% in width
6) column right menue dp25 (the pictures)
inside 6
6.1 box pic1
6.2 box pic2
...
7) column footer dp100 is same width of container
contains a box FooterTiddler
