Welcome to the TW world.

I am using Chrome 4.1.2... for test cases.

Edit the chorme windows link properties and add the --enable-file-
cookies parameter
like:
X:\..long path to exe..\chrome.exe --enable-file-cookies

Chrome will save cookies then to keep the local settings.

If you open the file, there should be the right main menu with:

close all
permaview
new tiddler
new journal
save changes
options »

If there is only

close all
permaview
options »

something went wrong.
