I am experimenting with TW5, TiddlySpot, Dropbox, and Chrome to work on and offline. No problem.
Make sure you input the TiddlySpot password before you save when uploading. Remember to change the name of the file when downloading. Firefox does not work smoothly with TW5 (and Chrome does not work smoothly with TWC.) For uploading TWC to TiddlySpot I use a portable version of Firefox version 3.6.
