VFB_DEFAULT_DEPTH is defined as 16 in xvnc.cc. It seems it has been defined as 16 in all RealVNC 4.X versions, see:

The define VFB_DEFAULT_DEPTH actually comes from Xvfb, which has been used as a template for Xvnc. In the 1.5.3 version of Xvfb, however, VFB_DEFAULT_DEPTH is defined as 8.
