Pushing the little button on the back actually does a reboot. The power
button on the screen does two things, it stops the player if its
playing any music and it activates the "off" screensaver (which you can
choose in the menus), thats it. It doesn't actually power down anything.
The server is still running so there should be no problem "waking up",
which just changes the screensaver! 

The only issue is how the drive behaves. Some drives have a sleep mode
which will spin down the drive if not being used for a certain period
of time. If this happens it might take some time for the drive to spin
back up and be ready to play music. 

There also seems to be some drives that actually disconnect themselves
from the USB port after a certain time, this is bad because the server
will then stop. If that happens you either have to plug and unplug the
drive or manually restart the server, in either case you then have to
wait for the rescan. 

So the big issue is to see how your drive responds after not being
accessed for a long time. The easiest way is to just don't play
anything over night, then check the status of the server, if its still
running then you should be fine. If the server isn't running it
probably means the drive has disconnected itself. The best solution for
that is to find some way to change its settings so it doesn't do that.
