I think the documentation is pretty clear here. In the second paragraph of the page you linked to (http://trac.edgewall.org/wiki/ TracPermissions ) it says:
"Permission privileges are managed using either the trac-admin tool from the command line or the new (in version 0.11) Admin tab."
