I've got no particular problem with the toolbar being on the left but it does put it quite far from the active areas which are the message area and the mailbox list area.
The toolbar will remain movable by the user. What I'm considering is moving its default position to the left, and maybe only in the wide layout ("three columns").
I also don't like the column layout. As per your screenshot, it makes the subject and from lines almost unreadable.
I got to resize the window a bit in order to make it small enough so that I don't feel bad sending a huge image by mail :).
I switched to using the wide layout on all of my screens (27" 2560x1440, 30" 2560x1600, and 14" 1600x900). On the 14" one, I usually use Trojita maximized, and on the rest of them, I typically have a window size of roughly 1600x1000px. That works well for me.
Having no text on the reply button is also a pain because I like to know if it's "reply all" or "reply one" before I click it and without waiting for the tool tip.
Trojita respects your system's settings here, so you should be able to configure the Qt style to show captions. There are also user-configurable shortcuts for various replying modes (hopefully with sane defaults), and you can change the mode of replies from the composer window. I hope this gets you reasonably covered :).
