Mary Alice committed suicide after years of accumulated guilt of killing her
son's birth mother.
The mother had been a drug addict living with the childless Paul and Mary
Alice; she had offered to sell the baby, but nearly OD'd before completing
the transaction. In the interim, they took the child, and moved out of town
and on to Wisteria Lane.
Several years later, the mother tracked them down and wanted her son back.
Mother-bear instincts kicked in on both sides; the birth mother knocked Paul
unconscious and went for her son, but Mary Alice took a knife and stabbed
her. (The knife -- I think, since I can't remember and Wikipedia isn't
refreshing my memory -- is what was in the box.)

up with the plumber?
If by "hook up" you mean "married," then your answer is the end of season 3.
(This was after his amensia, several ex-fiancé[e]s, kidnappings, yada yada
yada. Btw, the plumber is the bio dad from #1.)
