Not currently. In fact, Mr. Jobs went out of his way to note that it won't be the only way to get apps.
But once traction gets hold and the marketing machine ramps up, it'll be clearly known as the only SAFE place to get apps, the only APPROVED place to get apps, the only DESIRABLE place to get apps.
After that meme takes hold, it won't need to be mandatory. Other alternatives will just fade away.
Buying apps from a web site, which has been just fine as the conduit between loyal Mac users and loyal Max developers for more than a decade, will be seen as some sort of ghetto experience.
