Well, you can always check the AAAA (or A6) dns lookup for the address. But IPv6 is, unless a 6to4, a completely different address from the IPv4 address, and on a completely different routing schema, and to my knowledge there is no port or similar open to confirm active IPv6 on an IPv4 address. Finding the hostname and looking up the AAAA record is the only thing that comes to mind.
