I regularly lay out pamphlets and want to know if it is possible to layout pages in open-office so that Page 1 is opposite Page 16, for instance, and Page 2 oposite Page 15, etc.

Yes, it is possible, and it is done at the print stage. From the Print dialogue, click on Options, then select the Brochure option.

Currently I lay out in landscape using 2 columns in sequence, and then have to cut the pages apart and re-paste them. Is it possible to lay them out directly in Open Office?

It is not necessary to lay out the document the way you have. Just use normal A4 or A5 pages, and OpenOffice will automatically adjust the page sizes and page numbers for you.
