Yes,

  VBoxManage modifyhd FILENAME --resize

will work. Note that you will also have to resize your guest partition(s)
as the above command will only make the physical medium bigger. To resize
the partition(s) you can use a recent Ubuntu Live CD (yes, it can deal with
NTFS partitions as well).
