During Additions installation we create a script which should be run automatically at the beginning of every X session to start various VBoxClient sub-processes (clipboard, dynamic resizing, seamless mode). In order to make sure that it is started, we create a link to a script which starts them in any of the following directories that exist on the guest: /etc/X11/xinit.d /etc/xdg/autostart /usr/share/autostart /etc/X11/Xsession.d /etc/X11/xinit/xinitrc.d Perhaps Knoppix does not recognise any of these (or perhaps something goes wrong in the installation before that). In any case, you can add /usr/bin/VBoxClient-all to your X session to be sure that the clipboard will be available automatically.
