The error message is saying your guest machine is inaccessible because not all of its media are accessible. In particular, the ISO in the optical drive, it says, is inaccessible because it's already in use by another machine. I don't know how you got into that state, did you copy or clone the guest or something? In any event, you shouldn't need that ISO in the optical drive in order to boot the guest ... Try ejecting it from the existing machine, removing it from the media library if necessary. If that doesn't work, try closing & re-opening VirtualBox, ensure there isn't already a machine running.
