As Daniel said, automatic backups only happen when all 3 criteria are satisfied, but you did mention manual backups. Those you can of course do any time and all that is needed is a WiFi connection, the phone does not have to be plugged in and obviously it can’t be locked since you have to go to Settings, iCloud, Backup and tap on “Backup Now”. If you are not connected to WiFi the backup now option is not available, i.e. it is dimmed and can’t be tapped.
