I found that Siri will define the word provided she understands your spelling annunciation by saying something like "Define the word" and then slowly spelling the desired word. If you say anything like define the word spelled", for me she interpreted this as part of the word and it didn't work. But I'm still running iOS 6.1.4 and at best, you can say Siri is flaky and not very consistent.
