Hi Don, No, she has to use her sisters email address and the sister has to make sure that Facetime is enabled on her iPad under settings. By default the email addressed she uses for her Apple Id is used for Facetime on an iPad or iPod Touch, but she can also add other email addresses in which case she can be contacted via any of these addresses.
I believe you said your wife wants to initiate the Facetime call so she will have to create a contact for her sister. In this case all that would be needed is her name and email address. Then your wife can either use SIRI to initiate the Facetime call or she can go to the Contacts app, tap on her sisters name and then tap on :"Facetime" or "Facetime Audio". Facetime Audio is only available if both are running iOS 7, in iOS 6 only Facetime video calls were supported, but I think you said they wanted to do a regular Facetime call which means including video.
