Yes, you could work with him and be a backup MX for him, and he could be
a backup for you. 

Instead of setting up a Forced route for *, do the following, givin that
you host mydomain1.com, mydomain2.com, and mydomain3.com, and he hosts
hisdomain1.com, hisdomain2.com, and hisdomain3.com

Add the following forced routes on his mail server:
*.mydomain1.com  port25, preference0, and put in the IP address of your
mail server.
*.mydomain2.com  port25, preference0, and put in the IP address of your
mail server.*.mydomain3.com  port25, preference0, and put in the IP
address of your mail server.
Then add his mail server as a lower priority in your DNS records.

Add the following forced routes on your mail server:
*.hisdomain1.com  port25, preference0, and put in the IP address of his
mail server.
*.hisdomain2.com  port25, preference0, and put in the IP address of his
mail server.*.hisdomain3.com  port25, preference0, and put in the IP
address of his mail server.
Then add your mail server as a lower priority in his DNS records.

Can a standard version of VOP be used for the backup relay? I have a
business partner who is also running VOP. Would we be able to set up POs
on our systems for each other with a lower priority MX and become backup
for each other?

You would probably just want to use a modusGate or VOPMail Relay version
to setup on an alternate server (other network) and just set a forced
route for * to send all mail to your primary server.  Make this your
lower priority (higher number) mx record in DNS so if your primary is
offline, this server will queue mail for delivery to your primary when
its back online.
