I think reducing the number of colors without reducing the pixel size 
(bit per pixel) will reduce the image size anyway due to compression.

33333333 will compress better than 33333343 in GIF and many other 
format.

But as usual it's better to try than to guess. (I think most of you 
know this rule really well).
Compression is a very difficult topic. There are some few rules on 
image type, number of colors, compression algorithm etc... that may 
help you to do some predictions. But if eliminating 1 color will 
reduce your image of 1% maybe it will be better not to kill it.
