It's a little bit complicated because
'\Content.IE5\'
is a name used in several places.

Assuming this is a subdirectory of
\Temporary Internet Files\
then it should SHOULD only contain files used for Internet browsing and OE
news server message combining/browsing

So - if you are not worried about losing cookies as well as the files that
prior web and news server usage have created for the downloaded pages etc.,
then yes - you can just delete the contents including those subdirectories.

But before you do that consider that this particular occurrence of
'\Content.IE5\'
could be within your personal userid area (Documents and
Settings\.....\....\.)
or within a directory structure used for all users

It could hold very little, or lots depending on the actual usage.

Me, I only delete the stuff that I recognise as being pointless, or file
types that I recognise
( html htm gif js pdf swf jpg - and all those 0 length files)
and then only providing their date/time of creation indicates that they are
not in current use:
I.E. They are older than 24 hours, and if I didn't restart my system today,
pre-dating my last boot

The thing is that I don't want to lose cookies, password things and anything
that relate to what I'm doing via the internet now
but there SHOULDN'T be any long term problems just deleting all of the
'\Content.IE5\' contents

Note the frequent use of SHOULD

Also - how many files are there in that particular subdirectory structure,
and how much space do they occupy, and how many are 0 length?
