I think the colleague will have XQuartz.app in the usual location, and an X11.app under /Applications/MacPorts, that is at least the case for me. I'm not convinced that it will be easy to be 100% certain that the same X11 server is launched automatically. In my case I unload the launchd plists that come with XQuartz and port:xorg-server (and even move them out of the way). Since I always have X11 running I prefer to launch the server of choice by hand, from the Dock. The additional benefit is that my DISPLAY variable is simply set to :0.0 instead of to some complicated formula.
