It depends. I have many computers for development and testing, they all
share the same logical drive layout
(via appropriate partitioning), but physical configuration is different.
On desktops I have 2 (or 3) physical drives but on laptop I have one.
Note that drive letters in Windows NT/XP/Vista/Seven can be manually
assigned to partitions
using Control Panel -> Administrative Tools -> Storage / Disk Management
You don't need to rely on automatic / default assignment.

Yes, generally speaking any application no matter if it is 32 bit or
64 bit can be installed anywhere,
of course assuming that setup programs gives the user the choice of
installation folder.
Myself, I am installing programs on disk "F:" . "C" is my system drive
and I only have Windows installed there and nothing more.
I have 32-bit AmiBroker installed into "F:\AmiBroker\" and 64-bit
AmiBroker installed in "F:\AmiBroker64\"

and wanting to utilise *more* than 4GB of memory. 64-bit version does
