Yes and no. It depends on what you're doing.  Some people will for example plug a headphone-to-headphone cable from their keyboard to their PC.  You're just going to have to figure out how to adjust the levels on whatever you're using.  If it's a virtual microphone, the same procedures as a physical one--what I mentioned above--should apply.
