We just spent a weekend fighting this exact issue. Server 2008 domain 
controllers limit the number of connections that can be made by a single 
account.  Check out this support article: http://support.microsoft.com/kb/949469

We had to enable verbose logging on the DCs to see the errors, but this was the 
root cause of our BB issues as well as a couple of other things.

We also reboot BES whenever we have to reboot any DC or Exchange Server. This 
is likely the fix you need if you are on Exchange 2003.

Try restarting Dispatcher service; we have this problem intermittently on our 
4.1 and 5.0 servers whenever GC/DCs are rebooted.  Basically the BES doesn't 
reconnect to the DC correctly; thwn you try to send a message, it does a global 
catalog lookup to validate your account, and that is what is faililng and 
causing the red x.

If you are using Exchange 2010, there is something called throttling that needs 
to be adjusted for message flow to work. Also check MAPI profile on BES server.

Any idea what updating the domain controllers to 2008 R2 could have broken?  I 
just get a red X now trying to send external BB messages.
