I have encountered this same problem now with a couple of the 6.5b1 builds this week, including the latest one (6-24). Everything seems to work OK, install, re-scan DB from empty (always best with beta software), all the music gets scanned and the playlists built.
Then whether I just play one track directly, or a whole shuffled playlist, the players (I have 3 in a synch group, at the moment) all show the correct track, but nothing plays. You can hit the Slip button and it will skip to the next track - and then, not play it. Deleting this installation and installing the latest 6.3 has me running again (albeit with some other problems I'm still working through this afternoon). But since I've seen this same behavior as you on a couple of different 6.5b1 builds this week, I thought I should chime in.
