There is no such rule. That is why the EDM has been signed by MPs from
all parties.

It is true that it can be good tactics to get MPs from as many parties
as possible to be in the first six if you are going for maximum
signatories. But MPs tend to stick with their own, and tend to ask
their mates for signatories at first.

But there are also MPs who refuse to sign all-party EDMs (and others
who think EDMs are a waste of time, and won't sign any).

And in any case a lot of what are called rules are simply conventions
- such as front benchers not signing EDMs. It is not always observed.
