Scope of the meeting:
In the last decade, structural biology at synchrotron radiation sources
has been principally dedicated to the field of macromolecular
crystallography. However, modern structural biology relies more and more
on complementary methods that are applied simultaneously. While the
X-ray structure provides a global near-atomic view of a macromolecule,
spectroscopic data yields information on electronic structure,
vibrational levels as well as inter-atomic distances with sub-Angstrom
resolution. Synchrotron radiation facilities must adapt to this
evolution, in particular since it became clear that intense X-ray beams
can induce multiple chemical processes in macromolecules that need to be
assessed “online”. Integrated approaches where macromolecular
crystallography is combined with X-ray or UV-visible absorption and
fluorescence as well as Raman spectroscopy are particularly relevant to
the field of metallo- or chromophore containing proteins that display
pronounced spectroscopic signatures and also tend to be quickly damaged
by the X-ray beam. The purpose of this workshop is to review the recent
developments of spectroscopic techniques complementary to X-ray
diffraction at synchrotron radiation facilities as well as the new
science that is starting to emerge from using such integrated approaches.
