Hi all, I am sorry for the non-CCP4 question. I am trying to express a trimeric membrane protein on the yeast surface. The transmembrane domain of the protein also serves as the trimerization domain. The ecto-domain of the protein can be expressed in yeast as monomer but the full-length protein with the transmembrane peptide failed expression by western blot. I am suspecting that yeast does not like the non-native transmembrane domain. So I am planning to replace the transmembrane domain with a transmembrane domain from a yeast trimeric membrane protein, which is able to drive trimerization of the fused protein on the yeast surface. I am wondering if anyone knows such a protein. Thank you very much.
