5400 should be fine for PLAYBACK of most media, but if you are doing video editing and production, something a little faster would be nice. Like 10,000 RPMs, not 7200. And if you are serving the files up to a bunch of other computers via a network, you might need the extra RPMs
Don't worry about partitions. The space you lose will be small, and the ease of a single drive is worth the cost in terms of space, IMHO. But, for the love of God, be smart in your directory naming conventions. Remember the total file name length limitations when storing deep and long path and file names. And plan your backup methods before you start. Can you recreate it if neccessary? Are you going to back it up? To CD, DVD, tape or another drive? If to CD, remember that the ISO-9660 standard allows much shorter filenames, and less of the ASCII char set is allowable. If you think you don't need to back it up because (for instance) it is a bunch of mp3s you ripped from your own cds, think of the number of hours it took to rip and catalogue the media. And then back it up.
