Here are some of my favorites: http://www.penny-arcade.com - "Penny Arcade". Hilarious, but often gaming centric (not a problem for me) don't miss the news log either. They often linkto little known web comics that are worth the visit.
http://www.dieselsweeties.com/ - "Diesel Sweeties". Brilliant. My faviorite line EVER in a comic. After the a couple has been arguing "can not!", "can too" for a while an onlooker explains: "He thinks that Batman could menstruate if he wated too." http://www.gamespy.com/comics/dorktower/ - "Dork Tower". Geeky fun. http://www.explodingdog.com/ - "Exploding Dog". Not really a comic, but well worth regular visits. http://students.washington.edu/durandal/shawisland/ - "Shaw Island". Often (slightly) twisted, generally episodic and genius. Don't let one strip decide for you: read the archives. http://www.pvponline.com/ - "PVP". Very episodic (read the archives) gaming-focused comic but good on many, many other levels. http://www.goats.com/ - "Goats: The Comic Strip". Well-defined episodic stories. Occasionally brilliant. http://www.nthzine.com/toons/pclips/ - "Partially Clips". Stories added to classic clip art - better than it sounds. http://www.angryflower.com/ - "Bob the Angry Flower". A little hit of miss, but more hits than misses. http://dynamic5.gamespy.com/~extralife//index.php - "Extra Life". Again hit and miss, but worth the time. That's about it for me... but I'm always looking for new ways to goof off at the office.
