The ANC is calculated from the CBC (complete blood count) which is the routine blood test that is ran every couple of weeks at first.  It is the number of WBC multiplied by the neutrophil count.  ANC is absolute neutrophil count.  It is the number that determines rather you need additional shots...i.e. Neupogen or Procrit...to stay on the Gleevec.  Our oncologist uses 1000 as the cutoff.  So far my daughter has always been above 1000 on her ANC.
I do not think your doctor's nurse is correct.  The FISH test is run infrequently.  The PCR is what needs to be run at 3 and 6 months in order to give a good prognosis as to how you are going to respond long term to the Gleevec.  (This is what our doctor at OSHU does.)
