Hi Shane,

VX-680 shows very good activity against T315I in the lab, indeed. It
is in trial for blast crisis at MDACC, hope it opens soon for other CML
phases.

Regarding the slide you mention, do you know when Dr. Shah did the
presentation? For example, ONO12380 has been shelved, AP23464 is no
longer being studied. The only reason I say this is that research is
going at a fast pace and articles and presentations coming out all the
time. In Oct 2005, Wyeth published the IC50 for SKI against T315I
which is 344nM and much lower than Dasatinib, Imatinib and Nilotinib.
Whether this will be enough to overcome the T315I mutation, it would
depend once human trials are under way and doctors know the MTD dose.
We will know soon enough from Phase I data of SKI-606. I am also
keenly looking forward to data from VX-680.

SKI-606 trials are open at MDACC for chronic phase patients resistant
to Gleevec and it is a Phase I dose-escalating study. VX-680 from
clinical trials website is open for CML in blast crisis at MDACC but I
am sure will soon open up for other phases.

Best Wishes,
Anjana
