For an example of a big Drupal site look at http://www.ecademy.com/

I'm only just starting out with the PHP option and and also want to know the best system to start with.
It looks like people have been quite happy with phpNuke/postNUke, Typo3, and Drupal, but I haven't yet seen a comparison of features....

Any takers?

Drupal is an option, http://drupal.org/ It's PHP and either MySQL or
PostgreSQL. It's GPL'd and has a very active developer community...
