As a novice to making electron density difference maps, I am curious about the
algorithm underlying the “make a difference map” option in Coot. My impression
is that the subtraction occurs in real space rather than at the level of
structure factors. Given the remark in the Coot manual that this can be done
regardless of differences in grid parameters I would assume Coot interpolates
to a finer underlying grid. Are my inferences correct? A real-space subtraction
would be quite interesting, since I am comparing data sets that are/can be
processed in different space groups.
