Are you using the default skin. If so please try the latest 6.5.1 as this should speed up the time to build the playlist which looks like it is the web page taking the longest time to build.
In addition to this, the database line lead me to suspect that something is causing the database to respond slowly - you could try disabling any virus checker you have as a temporary measure to see if this helps.
