No, that shouldn't be hurting it.  But this is really sounding like your
parameter RAM battery (the one that keeps all the settings intact when the
Duo is otherwise unpowered) is dead.

You should be able to feel the brightness/contrast buttons "engage".  If
you're not sure, place one hand behind the screen to support it, and then
push in with your index finger, gradually increasing the pressure.  Once you
try it, you'll see that you can feel when the buttons are engaged.

As far as sleep, this is a low power use mode that preserves the contents of
RAM.  The processor, screen, HD, etc. are all powered down.  Since RAM is
preserved intact, when you wake from sleep, your Duo is in exactly the state
in which you left it.
